摘要
目的:探讨葛根素对糖尿病肾病(DN)大鼠肾病变的保护作用及其机制。方法:以高脂饲料喂养联合低剂量链脲佐菌素(STZ)复制DN大鼠模型,动物分为正常对照组、模型组、3个葛根素治疗组、依那普利治疗组、消渴丸治疗组。8周后检测空腹血糖、尿素氮、血肌酐(SCr)、微量白蛋白/尿肌酐比值(U.A/C)和总蛋白/尿肌酐比值(UTP/C)等;Masson染色法观察肾组织形态学改变;免疫组织化学检测肾组织纤维连接蛋白(FN)与IV胶原蛋白(IV-C)的表达。结果:ig给药,葛根素可改善DN大鼠肾脏功能和肾组织形态学异常,降低肾组织中FN与IV-C的表达。结论:葛根素对DN大鼠肾脏具有保护作用,此作用可能与下调肾脏组织FN与IV-C的表达有关。
Objective: To investigate the effect of puerarin on diabetic nephropathy and the mechanism in rats. Methods: The rat model of diabetic nephropathy was induced by intraperitoneal injection of low-dose of streptozocin (STZ) after having high-fat diet for six weeks. The rats were treated with 3 doses of puerarin, enalapril or XiaoKe Wan every day. At 8 weeks after treatment, biochemical indicators were evaluated. The pathological changes in the kidney were determined by Masson staining. The expression of fibronectin (FN) and type IV collagen (IV-C) were detected by immunohistochemistry. Results: Puerarin suppressed histological changes in the kidney tissues, and reduced the levels of these biochemical parameters and the expression of FN and IV-C proteins in renal tissues of diabetic rats with nephropathy. Conclusion: Puerarin protects renal function and slows down the progression of diabetic nephropathy in rats by suppressing the expression of FN and IV-C proteins in renal tissue.
