摘要 目的观察小牛血清去蛋白注射液(Deproteinised calf blood serum injection,DCBSI)对原代培养皮层神经元树突发育是否有促进的作用。方法选用出生0~24 h的Sprague-Dawley(SD)大鼠乳鼠,取皮层神经元进行体外细胞培养,7 d后用于实验。应用倒置相差显微镜和微管相关蛋白-2(MAP-2)免疫荧光染色观察神经元树突的生长。结果 DCBSI(1,10,100μmol.L-1)可明显促进神经元树突发育,表现为神经元树突总长度明显增加、树突末梢分支数明显增多,并呈明显浓度依赖性。结论 DCBSI对体外培养皮层神经元树突发育有促进作用。 Objective To investigate the effects of deproteinised calf Blood serum injection on dendritic development in cultured cortical neurons. Method Cortical neurons cultures were prepared in vitro from postnatal Sprague - Dawley (SD) rats in 24 hours. After 7 days, cortical neurons were used for experiment. Neuronal dendrite outgrowth was observed by using inverted phase - contrast microscope and microtubule associated protein - 2 ( MAP - 2) immunofluorescence stai- ning. Results DCBSI (1, 10, 100μmol·L-1) can significantly promote neuronal dendritic development, such as the total length of dendrites of neurons increased significantly, dendrites branching number increased considerably, and in a dose dependent manner. Conclusion The results show that DCBSI can promote dendritic development and maturation of postnatal cortical neurons in vitro.
