重组巴斯德毕赤酵母Mut^s型与Mut^+型菌株产真菌α-淀粉酶发酵性能的比较 被引量:1

摘要 筛选了一株产真菌α-淀粉酶重组毕赤酵母甲醇慢速利用型菌株(Mut^s)KM71/pPIC9KRoAmy,然后在摇瓶水平较系统地比较了其与本实验室先前构建的甲醇快速利用型(Mut^+)GS115/pPIC9K-RoAmy之间发酵性能的区别,包括生长阶段和产酶阶段菌体生长情况以及最适pH,不同装液量、混合碳源流加对诱导产酶的影响,最后在15 L发酵罐上进行放大试验。通过对比发现,Mut^s表现出了较好的应用潜力。 For remitting the high demand of dissolve oxygen in the fermentation process of Mut^+ ( methanol utilization plus growth phase and fungal a-amylase production phase, different liquid medium volume, mixed carbon source fed-batch cul-ture, and finaUy scaling up in a 15 L fermenter, all of these indicated that Mut^s performed better than Mut^+ in fungal α- amylase expression in many ways.
