摘要
目的:体外诱导人脐带间充质干细胞(human umbilical cord mesenchymal stem cells,UCMSCs)向甲状腺细胞分化,探讨人UCMSCs向甲状腺细胞分化的可能性。方法:从胎儿脐带中分离UCMSCs培养至第三代,流式细胞仪鉴定后分为两组,诱导组接种于含胰岛素、促甲状腺激素因子的培养液中,对照组接种于完全培养基中。对细胞形态学进行观察;流式细胞仪鉴定诱导后细胞表面抗原TTF-1和TSHR的表达情况。结果:培养的P3UCMSCs呈长梭形、漩涡状生长,经鉴定表达间充质干细胞抗原;诱导组细胞形态d4开始发生变化,有些细胞变成三角形、不规则生长。对照组仍为长梭形;诱导后d12经检测诱导组TTF-1和TSHR的表达明显高于对照组。结论:人UCMSCs在特定的培养条件下有向甲状腺细胞分化的可能性。
Objective To investigate the feasibility of the differentiation of human umbilical cord mesenchymal stem cells (UCMSCs) into thyroid cells in1 vitro. Methods UCMSCs, which were isolated from fetal umbilical cord, were cultured to the 3rd generation, and then were divided into induction group and control group after being identified by flow cytometry. UCMSCs in induction group were seeded in culture fluid containing thyrotrophin and insulin, and those in control group were seeded in complete medium. Morphological changes were observed by the inverled optical microscope and flow cytometry was used to detect the expressions of surface antigen TTF-1 and TTF-1 in the differentiated human UCMSCs. Results The cultured UCMSCs grew in long spindle and swirling, and were identified to express mesenchymal stem cell antigen. At day 4, the morphology of UCMSCs in induction group was changed from spindle to round or triangle, and grew irregularly with clear boundau. At day 12, the expressions of TTF-1 and TTF-1 were higher in induction group than those in control group. Conclusion UCMSCs could be induced to differentiate into thyroid cells in vitro in certain condition.
