Expression and significance of TGF - β_1 and Smad 4 in cervical tissue

摘要 目的研究转化生长因子β1(transforming growth factor-beta1,TGF-β1)和Smad4在宫颈上皮内瘤变(cervical intraepithelial neoplasia,CIN)及ⅠA2-ⅡA宫颈鳞状细胞癌(squamous carcinoma of the cervix,SCC)中的表达,并探讨其与SCC患者临床病理资料之间的关系。方法采用免疫组化S-P法检测TGF-β1、Smad4在35例CIN、50例SCC、10例正常宫颈组织中的表达并分析其在肿瘤不同临床病理特征之间的关系。结果 TGF-β1蛋白在正常宫颈鳞状上皮组织、CIN、SCC组织中表达强度渐增加,3组之间比较,差异有统计学意义(P<0.05);且TGF-β1在SCC中的表达强度与SCC的病理分级、有无淋巴结转移、浸润深度密切相关(P<0.05)。Smad4蛋白在正常宫颈鳞状上皮组织、CIN、SCC组织中表达强度渐减弱,3组之间比较,差异有统计学意义(P<0.05),且Smad4在SCC中的表达强度与SCC的病理分级、有无淋巴结转移密切相关(P<0.05)。TGF-β1和Smad4在SCC中的表达呈负相关性(P<0.05)。结论 TGF-β1、Smad4蛋白可能在SCC发生发展以及转移方面发挥了一定的作用,它们的表达与SCC病理分级有关,有望作为判断SCC恶性程度的指标。 Objective To investigate the expression of transforming growth factor - beta 1 ( TGF - 131 ) and Smad 4 in cervical intraepithelial neoplasia(CIN) , squamous carcinoma of cervix(SCC) and the clinical significance, and investigate their relationship in different clinical characteristic of neoplasm. Methods The immunohistochemical method was used to detect the expression of TGF - 131 and Smad 4 in 35 cases with CIN,50 cases with SCC and 10 cases with normal cervical epithelium tissue paraffin sections, and investigate the relationship in different pathological character. Results The expression levels of TGF - 13, increased with increasing severity of cervical disease ( P 〈 0.05) ,TGF- 13t had closely relative to pathological differentiation, infiltration and lymph- node metastases of squamous carcinoma of the cervix ( SCC ) and the difference was significant ( P 〈 0. 05 ). Smad 4 expression level decreased with increasing severity of cervical disease ( P 〈 0.05 ) , and Smad 4 had closely relative to pathological differentiation and lymph - node metastases of SCC ( P 〈 0.05 ). The expression of TGF -β1and Smad 4 had apositive correlation in SCC. Conclusion TGF - ~, and Smad 4 may play a certain role in SCC development and the transfer, their expression related to SCC pathological grade, they are expected as an indicator to determine the degree of malignancy of SCC.
