摘要
建立了高效液相色谱法测定麻保沙星溶液含量的方法。以23%甲醇溶液溶解并稀释成每1 mL中含麻保沙星0.1 mg的溶液作为供试品,用十八烷基硅烷键合硅胶色谱柱,以醋酸铵高氯酸钠溶液-乙腈(85∶15,V/V)为流动相,检测波长291 nm,流速1.0 mL/min,柱温40℃,进样量10μL。结果表明,麻保沙星溶液在5~200μg/mL范围内线性良好,线性方程为Y=4.6×104X+3.64×105,r=0.9998;平均回收率为98.3%~100.3%,批内变异系数为0.4%~1.0%,批间变异系数为0.4%~0.9%。本方法准确、可靠,适用于麻保沙星溶液的含量测定。
To establish a high performance liquid chromatography method for determination of the content of marbofloxacin solution. The sample dissolved with 23% methanol solution and diluted to 0.1 mg/mL, and then assayed by HPLC. The separation used with ODS chromatography column, and ammonium acetate/sodium perehlorate solution and acetonitrile (85 : 15, V/V) was used as mobile phase. The detection wavelength was 291 nm, the flow rate was 1 mL/min, and the column temperature was 40 ℃. The linear equation was Y=4.6×10^4X+3.64×10^5, and correlation coefficient was r = 0. 9998, as marbofloxacin at 5 - 200 μg/mL. The average recoveries were 98.3% - 100.3% , the intra - assay coefficient of variation were 0.4% - 1.0%, and the inter - assay coefficients of variation were 0.4% - 0.9%. The results show that, this method is accurate, reliable and applicable to the determination of content of marbofloxacin solution.
