Role of c-kit in the pathogenesis of intestinal hypomotility induced by anthraquinones laxatives

摘要 目的探讨c-kit在蒽醌类泻剂所致肠动力减退发病机制中的作用。方法健康成年SPF级SD大鼠36只随机均分为模型组、模型恢复组和对照组。模型组和模型恢复组大鼠采用"大黄酸混悬液灌胃法"制作肠动力减退动物模型,对照组用等体积生理盐水灌胃。模型组造模结束后取材,模型恢复组造模结束正常饲养30d后取材。各组大鼠处死前观察首粒黑便排出时间以检测肠道传输功能,Western blot法检测各组大鼠结肠组织中c-kit的蛋白水平。结果模型组和模型恢复组大鼠首粒黑便排出时间较对照组明显延长(P<0.01);模型组和模型恢复组结肠组织中的c-kit蛋白表达较对照组明显减弱(P<0.01)。结论蒽醌类泻剂所致肠动力减退可能与结肠组织中c-kit蛋白表达下调有关。 Objective To investigate the role of c-kit in the pathogenesis of intestinal hypomotility induced by anthraquinones laxatives. Methods Thirty-six healthy adult SPF SD rats were equally randomized into three groups. The rats in groups of A and B were given rhein suspension by garage to produce the models of intestinal hypomotility,which in group C were given same volume of saline by gavage as the controls. The rats in group A were sacrificed for detecting the expression of c-kit protein in colon tissues by Western blot after the models were successfully established, while those in group B were fed for 30 days and harvested thereafter. The intestinal transit function was detected through observing the first time of defecation before the rats were killed. Results The first time of defecation was longer in groups of A and B than that in group C(P〈0. 01). The expression of c-kit protein was lower in groups of A and B than that in group C (P〈0. 01 ). Conclusion The intestinal hypomotility has not recovered after anthraquinones laxatives withdrawal, which may be associated with downregulation of c-kit protein expression in colon tissues.
