摘要 目的研究肿瘤转移相关基因1(MTA-1)与肿瘤转移抑制基因(KISS-1)表达蛋白在膀胱癌中的表达及临床病理意义。方法采用免疫组化SP法检测65例膀胱癌组织和15例膀胱正常组织中MTA-1和KISS-1的表达,结合临床资料及组织病理学特点探讨MTA-1和KISS-1基因表达与膀胱癌浸润和转移的关系。结果 MTA-1蛋白在膀胱癌组织阳性表达率明显高于膀胱正常组织(53.85%vs.6.67%)(P<0.01);KISS-1蛋白在膀胱癌组织阳性表达率明显低于膀胱正常组织(40.00%vs.86.67%)(P<0.01);其基因表达与病理分级、临床分期、淋巴结转移密切相关(P<0.05)。膀胱癌组织中MTA-1与KISS-1蛋白表达呈负相关(P<0.01)。结论 MTA-1蛋白高表达和KISS-1蛋白低表达与膀胱癌的浸润、转移密切相关,可作为诊断膀胱癌患者转移、复发和判断预后的重要参考指标,有望成为膀胱癌基因治疗的新靶点。 Objective To study the expression and clinicopathological significance of metastasis-associated gene I(MTA-1) and tumormetastasis KISS-1 in bladder cancer. Methods The expressions of MTA-1 and Kiss-1 were detected by immunohistochemical SP technique in 65 cases with bladder cancer (group A) and 15 normal controls (group B). The association between the expressions of MTA-1 and Kiss-1 and clinicaopathological parameters was analyzed. Results The positive rate of MTA-1 was higher in group A than that in group 13(53.85% vs. 6.67%)(P〈0. 01). The positive rate of KISS-1 was lower in group A than that in group B(40.00% vs. 86.67%) (P〈0. 01). The expressions of MTA-1 and Kiss-1 were closely related to the TNM stage, infiltration and metastas of bladder cancer(P〈0. 05). The expression of MTA-1 was negatively correlated to that of KISS-1 in bladder cancer(P〈0. 01). Conclusion High expression of MTA-1 and low expression KISS-1 are significantly involved in the infiltration and metastas of bladder cancer. Detection of MTA-1 and KISS-1 in the bladder cancer may be useful markers for predicting the recurrence, metastasis and prognosis in the patients with bladder cancer, which is likely to be a new target of gene therapy of bladder cancer.
