摘要
在正己烷中应用工业用脂肪酶催化油酸和正十二醇的酯合成反应 ,研究了该反应的动力学机制 ,考察了酶和底物浓度对反应初速度的影响。结果发现 :反应过程中不存在底物抑制及扩散限制 ,反应为动力学控制 ,符合Michaelic -Menton方程。脂肪酶催化动力学符合Ping
The kinetics of lipase-catalyzed esterification of oleic acid and dodecanol in hexane was investigated. The effect of enzyme and substrate concentration on the initial rate of esterification was studied, and the reaction kinetics was described in terms of the Michaelis-Menton mechanism. No evidence of inhibition by oleic acid and dodecanol was found at all substrate concentrations. The kinetics constants were measured for the reaction in the absence of any significant diffusional limitations.It was suggested that the kinetics of the enzymatic reaction agreed with a Ping-Pong Bi Bi mechanism.
