红景天苷干预小鼠肝纤维化的研究 被引量:12

Effect of Rhodioside on the Progress of Liver Fibrosis in Mice

摘要 目的:以药物诱导的小鼠肝纤维化为实验模型,研究中药红景天苷对该模型是否有治疗作用,及可能的作用方式,结合该药对大鼠肝纤维化的治疗作用,为临床应用该药提供指导。方法:选取10对同窝同性别8周龄小鼠,每对内两只小鼠随机分进实验组(experimental group,EG)与对照组(control group,CG)。进行CCl4腹腔注射诱导小鼠肝纤维化,并在诱导中期实验组加入红景天苷,对照组继续原诱导方案。诱导12周结束后取小鼠肝组织进行H&E染色、MASSON染色、巨噬细胞的免疫荧光染色及血清Hyp含量表达测定。结果:实验组小鼠肝组织形态较好,假小叶形成较少,胶原沉积也相对较少(P<0.001),其血清中的Hyp含量低于对照组(P<0.01),以上差异有统计学意义。在实验组小鼠肝脏中,F4-80标记的巨噬细胞百分比明显低于对照组(P<0.001),该差异有统计学意义。结论:红景天苷能够影响小鼠的正常肝纤维化进程,该药减轻了小鼠的肝纤维化程度,对小鼠肝纤维化进程有一定的保护作用。此作用的发挥可能是该药物影响了肝脏中的Kupffer细胞的增殖或者凋亡,从而使得该细胞对炎性信号调控发生变化,使得肝星形细胞分泌胶原减少得以实现。该药在大鼠及小鼠肝纤维化模型上的治疗作用提示其临床应用前景较好。 Objective: In order to guide the clinical medication of Rhodioside. According to its effect on rats, we investigated the therapy effect of Rhodioside on liver fibrosis of mouse which induced by carbon tetrachloride, and we found out the potential type of the effect in this model. Methods: Ten pairs of mice was used and each pair was randomly sub-divided into EG or CG group. Each pair was from the same brood and they were of the same sex and all at the age of eight weeks. We used the carbon tetrachloride with the method of peritoneal injection to induce mouse liver fibrosis. In the metaphase, Rhodioside was added into the EG. Twelve weeks later, liver fibrosis was detected by using H&E and Masson pigmentation and the level of Hyp in mouse blood serum. Immunofluorescence was used to detect the percentage of macrophage in liver. Results: In the experimental group, mouse liver has a better hepatic structure, pseudo lobule and the depositive collagen fiber is not that much as the control group (P〈0.001), and the level of HYP in the blood of EG is significantly lower than CG(P〈0.0I), the F4-80 labeled macrophage percentage is much lower in the EG compared to CG(P〈0.001). All of the indexes above have significant deviation. Conclusion: Rhodioside has effects on liver fibrosis progress of mouse; it can lighten the degree of mouse liver fibrosis, so it has protection effect. This action may depend on Rhodioside influenced the Kupffer cell's generation or apoptosis which regulated the inflammation signaling pathway, and which can down regulated the collagen which secreted by stellate cell. Owe to its good therapy effect on rats and mouse, this medicine has good prospect on clinical medication.
