摘要 为研究TEAD1和YAP1对牛IFNT基因的表达调控,利用PCR分析检测牛胚胎及各脏器中TEAD1和YAP1的表达情况。并通过IFNT荧光素酶报告基因,分析TEAD1和YAP1对牛IFNT基因在JEG3细胞中表达的影响。结果显示,TEAD1和YAP1基因在牛着床前胚胎中及各脏器中都有表达。荧光素酶分析表明,在IFNT基因5′端上游区域逐渐切除或者点突变的情况下,YAP1对IFNT基因的表达都有显著的上调作用;TEAD1对IFNT基因表达的影响不显著;TEAD1和YAP1共同作用时,协同效果不显著。 In order to study the transcriptional regulation of TEAD1 and YAP1 on bovine IFNT gene expression, PCR analysis was performed to detect the expression of TEAD family and YAP1 in bovine concepus and organs. Through the IFNT Luciferase reporter gene analysis, effect of TEAD1 and YAP1 on IFNT gene expression was studied in JEG3 cells. The result indicated that TEAD1 and YAP 1 gene could be expressed commonly in bovine concepus and organs. YAP1 significantly up-regulated the IFNT gene expression (P〈0.05), and 5'-deletion could not impair this up-regulation; TEAD1 had no significant effect on IFNT gene expression~ There was no significant synergistic effect between TEAD1 and YAP1.
