摘要
本研究在非水介质中酶法合成了氨基酸神经四肽Phe-Arg-Trp-Gly(FRWG)的前体二肽BOC-Phe-Arg-OMe,胰凝乳蛋白酶(ChyT)作为催化剂在水-有机溶剂体系中催化合成目标二肽.通过考查有机溶剂种类、水含量、温度、pH等对产物二肽BOC-Phe-Arg-OMe的影响,优化了合成反应的条件.并确定了最佳反应条件:使用胰凝乳蛋白酶作为催化剂,在pH为7.5,温度40℃,二甲基甲酰胺(DMF)/Tris-HCl缓冲液(98.5 1.5,V/V),反应8 h,二肽产量达到61.25%.
The protease-catalyzed synthesis of a precursor dipeptide of neural tetrapeptide ( Phe-Arg-Trp-Gly), BOC-Phe-Arg-OMe in non-aqueous media was studied. An chymotrypsin was used to catalyze the synthesis of the target dipeptide in water-organic cosolvents systems. The conditions of the synthesis reaction were optimized by examining the effects of several factors including organic solvents, water content, temperature, and pH in DMF/Tris-HCl buffer system (98.5 : 1.5 ,V/V) ,8h with the dipeptide yield of 61.25%.
