Purification of seabuckthorn flavonoids by metal complex method

摘要 研究金属络合法纯化大果沙棘黄酮的工艺。对6种不同金属与大果沙棘黄酮的络合效果进行了比较,并通过单因素试验优化金属络合法纯化大果沙棘黄酮的最佳工艺。筛选出最佳络合金属为乙酸锌,得到金属络合法纯化大果沙棘黄酮的最佳工艺条件为:黄酮浓度为0.20mg/mL、络合时间30min、乙酸锌溶液浓度6.9mmol/L、反应液pH值为8.0、解络剂EDTA用量为1.2g/g黄酮。在此纯化条件下,黄酮纯度由粗提物的9.1%增加到52.1%,提高了5.8倍。 The process parameters of the purification of seabuckthorn flavonoids by metal complex method were studied. Six different kinds of metal were studied on the complex capacity with flavonoids. The optimal process for the purification of flavonoids was explored on the basis of single factor experiments. The best metal was zinc acetate. The optimized technology conditions to purify seabuckthorn flavonoids were as follow: flavonoids concentration was 0.20 mg/mL, time 30 min, zinc acetate solution concentration was 6.9 mmol/L, pH value was at 8.0, EDTA was 1.2 g/g flavonoids. After being purified, the purity was increased from 9.1% to 52.1% with 5.8 times as high as the crude extracts.
