雷帕霉素对大鼠慢性肾间质纤维化中HGF、TGF-β的表达的影响 被引量:7

摘要 目的:观察雷帕霉素对大鼠肾间质纤维化的干预作用并探讨其可能的作用机制。方法:将Wistar雄性大鼠随机分为3组,即NS组(n=8)、模型组(n=24),治疗组(n=24),每天予以200mg/kg的腺嘌呤灌胃,连续喂养6周,在第2、4、6周采集大鼠的血和尿,测定血肌酐、白蛋白和24h尿蛋白,并取出肾组织,采用Masson染色法观察各组肾间质纤维化的程度,免疫组化法分析大鼠肾组织中HGF、TGF-β的表达情况。结果:治疗组的大鼠24h尿蛋白及血肌酐值明显低于模型组(P<0.05),白蛋白水平高于模型组(P<0.05),与模型组比较治疗组的肾间质组织中HGF的表达水平显著升高(P<0.05),NGAL的表达水平显著升高(P<0.05)、TGF-β的表达水平显著降低(P<0.05)。结论:雷帕霉素能降低24h尿蛋白和血肌酐的含量并能升高白蛋白的水平,提高HGF和降低TGF-β在大鼠肾纤维化组织中的表达,从而能够减轻肾纤维化的的程度,达到保护肾功能的作用。 Objective: To observe the interventional effect of rapamycin on rats with renal interstitial fibro- sis and to explore its potential mechanism. Methods: The male Wistar were randomly divided into 3 Groups: NS Group (n=8), model group (n=24), and treatment group (n=24), receiving 200mg/kg of intragastrie amidopurine daily for 6 weeks. On the 2nd, 4th, and 6th weeks, the blood and urine of the Wistar were collected, serum ereatinine, albumin and 24h urinary protein were all determined. And then the kidney tissues of all groups were taken out to observe the degree of renal interstitial fibrosis by means of Masson staining method, and to analyze the expression of HGF, and TGF--beta in kidney tissue by means of Immunohis- tochemical method. Results: 24h urinary protein and serum creatinine of treatment group was obviously lower than that of model group (P 〈 0.05), albumin levels higher than that of the model group (P 〈 0.05). Com- pared with model group, the expression of HGF in renal interstitial tissue of the treatment Group was signifi- cantly higher (P 〈0.05), expression level of NGAL increased (P 〈 0.05), and expression of TGF-β signifi- cantly lowered (P 〈0.05). Conclusion:Rapamycin can reduce 24h urinary protein and serum creatinine levels, increase levels of albumin and HGF, and decrease TGF-β expression in rat renal fibrosis so as to reduce the level of renal fibrosis and protect renal function.
