IL-5单克隆抗体对bcl-2和bax在鼻息肉中表达的影响 被引量:1

摘要 目的:研究IL-5细胞因子对于鼻息肉组织的影响及其作用机制。方法:①24例鼻息肉患者,男女各半。其组织标本均在局麻下鼻息肉摘除手术中获得。将每份鼻息肉标本分为空白组(KB)、单抗(DK)组(年龄14~58岁,按1997年海口标准)分别培养1、3、5、7天,每时间点各6例标本。②用在体外组织块培养、免疫组织化学法、原位末端标记的方法在体外模拟体内环境中观察IL-5单克隆抗体对于鼻息肉组织中的bcl-2、bax表达的影响。结果:①IL-5单克隆抗体能够抑制鼻息肉组织中的IL-5表达(P<0.01),减少了鼻息肉组织的EOS浸润,促进了息肉组织的细胞凋亡。②IL-5在鼻息肉组织中第一天就有表达,第三天表达出现高峰,而bcl-2蛋白的表达却相对落后,在培养第5天才达到峰值,且延续时间较长。③在单克隆抗体组bax蛋白的表达较IL-5的表达相对落后在第5天才达到峰值。④单克隆抗体组鼻息肉组织在第一天就有凋亡细胞的表达在第5天达到峰值,第7天时仍未降至正常,均显著高于空白组。结论:①IL-5单克隆抗体可以显著抑制IL-5在鼻息肉组织中的表达。②IL-5细胞因子通过打破了bcl-2/bax的表达平衡,激活或增强bcl-2在鼻息肉组织表达延缓了鼻息肉组织凋亡时间是IL-5引起嗜酸性粒细胞浸润增多的主要原因之一。③IL-5的表达可以抑制或减弱鼻息肉组织的凋亡细胞的表达,延长鼻息肉组织的生存时间。 Objective: To explore the effect and mechanism of Interleuk--5 on the nasal polyp tissue. Methods.(1)we collected 24 patients with nasal polyp tissues. Those nasal polyp tissues were divided into mono- clonal antibody group (DK) and control group (KB). The time (1st day, 3rd day, 5th day, and 7th day) was established for two groups. (2)Immunohistochemistry and a terminal deoxynucleotidyl transferase mediated d-- UTP--floursceinend--labeling (TUNEL) assay were applied to observe the effect of Interleuk--5 monoclonal antibody treatment on the expression of bcl--2 and bax in nasal polyp tissues. Results: (1)Interleuk--5 mono- clonal antibody treatment inhibited the expression of Interleuk--5 positive cells, and aggravated the apoptosis of nasal polyp tissues. (2)The expression of Interleuk--5 protein positive cells occurred on the first day, the peak was found at the third day, and meanwhile, the peak expression of bcl--2 positive cells occurred on the fifth day but was observed in the same reign. (3)The expression of TUNEL positive cells occurred on the first day, the peak was found on the fifth day, and the positive cells of monoclonal antibody group were much more than control group. (4)The expression of bax positive cells occurred on the first day; the peak was founded on the fifth day, and the positive cells of monoclonal antibody group were much more than control group. Conclusion: (1)Interleuk--5 monoclonal antibody treatment inhibited the expression of Interleuk--5. (2)It suggests that In- terleuk--5 broke the expression balance between bcl--2 and bax, and their expression may be modulated partly by Interleuk--5expression and Interleuk--5 may protect the nasal polyp tissues from apoptosis thereby. (3)Inter- leuk--5 monoclonal antibody treatment decreased the cell apoptosis in nasal polyp tissues.
