摘要
利用rDNA ITS序列分析结合真菌形态学和生理学对兔的病原菌进行鉴定。提取菌株DNA,利用引物ITS1和ITS4进行PCR扩增,扩增产物测序后在GenBank核酸数据库中进行同源性比对。结果显示,分离株的菌落形态为白色粉末状,背面有色素沉着,培养可见大量葡萄状小分生孢子,螺旋菌丝及棒状大分生孢子;尿素酶和毛发穿孔试验阳性;rDNA序列比对结果显示,该分离菌株与须癣毛癣菌复合体中的万博节皮菌有99.8%同源性;从形态学和生理学试验符合须癣毛癣菌的特点,结合分子生物学试验将该菌株鉴定为万博节皮菌。
The fungal pathogens of rabbits were identified by using rDNA ITS sequence analysis combined with morphology and physiology.The DNA of strains was extracted for PCR amplification using primers ITS1 and ITS4.The amplified products were sequenced and blasted in Genbank for homology comparison.Results showed that the clinical isolates yielded white and powdery colonies with yellow reverse pigmentation.The fungal cultures revealed numerous grape-like microconidia,spiral hyphae and club-shaped macroconidia.Urease test and hair perforation test were positive.The sequences of this isolates showed 99.8% homology with Arthroderma vanbreuseghemii in Trichophyton mentagrophytes complex.The morphology and physiology of tested strains met the characteristics of Trichophyton mentagrophytes.The strains were identified as Arthroderma vanbreuseghemii by rDNA ITS sequence analysis combined with morphology and physiology.
