摘要
采用Ⅳ型胶原酶灌流法分离鸡肝细胞进行原代培养,用MTT法测定试验鸡肝细胞的存活情况,根据OD值计算药物对鸡肝细胞的生长抑制率;用CCl4体外诱导肝细胞损伤模型,检测培养上清液中天门冬氨酸转移酶(AST)和丙氨酸氨基转移酶(ALT)水平以及细胞形态学观察,比较各药物对肝细胞损伤的保护作用。结果表明,葡萄糖醛酸内酯、促肝细胞生长素、联苯双酯、肝泰乐、白芍的毒性较强,黄芪、茯苓、大黄、白术的毒性较小;体外肝细胞损伤保护试验结果表明,柴胡、垂盆草、当归、大黄、北五味子、黄芩、华蟾素、促肝细胞生长素等药物能明显降低ALT水平,与CCl4损伤组相比差异极显著(P<0.01),对AST也有较明显的降低作用,与正常组相比差异极显著(P<0.01),白芍、决明子、甘草、茯苓、丹参、黄芪、三七、肝泰乐、联苯双酯、葡萄糖醛酸内酯组对降低AST,ALT也具有一定的作用。柴胡、黄芩、垂盆草、北五味子、丹参、益肝灵片、葡萄糖醛酸内酯、甘草、黄芪对鸡肝细胞毒性较低,对CCl4引起的鸡肝细胞损伤具有一定的保护作用。
The primary chicken hepatocytes were isolated by perfusion with IV-type collagenase.Measurement of trititaed thymidine(MTT) was applied to determine survival cell information in the primary chicken hepatocyte test,the growth inhibiting rates of the medicines to the healthy cells were calculated with the optical density(OD) value.The primary chicken hepatocytes were injured by CCl4.The levels of AST and ALT in cultural supernatant were determined by general methods to observe the function of protection of hepatocytes.The toxicities of glucurolactone,hepatocyte growth-promoting factors,biphenyl dimethylester,glucuronic acid lactone and Paeonia sterniana to the cells were higher than that of others.The Astragalus membranaceus,Poria cocos,Rheum palmatum and Atractylodes macrocephala had smaller toxicity than others.Bupleurum chinense,Sedi sarmentosi,Angelicae sinensis,Rheum palmatum,Fructus schisandrae,Astragalus membranaceus,Chinese bufonin and hepatocyte growth-promoting factors can significantly degraded the levels of AST,ALT(P0.01).The other groups of Paeonia sterniana,Catsiatora,Glycyrrhizae,Poria cocos,Salvia miltiorrhiza,Astragalus membranaceus,Panax notoginseng,glucurolactone,biphenyl dimethylester,glucuronic acid lactone can also degrade the levels of AST and ALT in cultural supernatant.Bupleurum chinense,Scutellaria baicalensis,Sedi sarmentosi,Fructus schisandrae,Salviae miltiorrhizae,glucurolactone,Wolfiporia cocos,and Astragalus membranaceus had lower toxicity and could protect hepatocytes from in vitro injury induced by CCl4.
