摘要
目的探讨姜黄素对大鼠肾缺血再灌注损伤(ischemia and reperfusion injury,IRI)组织TLR4表达的影响。方法将36只大鼠随机分为假手术组、缺血再灌注组和姜黄素组,每组各12只,均切除右肾,游离左肾,假手术组直接缝合腹壁,余两组行肾脏IRI实验,其中姜黄素组试验前予姜黄素。3组均检测血肌酐(Cr)、尿素(BUN),光镜下观察肾组织病理学改变,Western blotting法检测TLR4蛋白表达,并进行比较。结果缺血再灌注组和姜黄素组Cr和BUN水平均明显高于假手术组,差异有统计学意义(P<0.05);与缺血再灌注组比较,姜黄素组的Cr和BUN水平降低,差异有统计学意义(P<0.05)。假手术组、姜黄素组光镜下见肾小管上皮细胞无明显改变,而缺血再灌注组光镜下见肾小管上皮细胞明显变性坏死。假手术组大鼠肾组织内有少量的TLR4蛋白表达,而缺血再灌注组和姜黄素组的TLR4水平明显高于假手术组,差异有统计学意义(P<0.05);与缺血再灌注组比较,姜黄素组TLR4表达水平显著降低,差异有统计学意义(P<0.05)。结论姜黄素能减轻肾脏IRI,其机制可能是通过抑制TLR4的表达而发挥作用。
Objective To investigate effects of curcumin on expression of toll-like receptor 4 (TLR4) in renal ischemiareperfusion injury (IRI) in rats. Methods A total of 36 rats were randomly divided into sham operation group (n = 12), ischemiareperfusion group (n = 12) and curcumin group (n = 12). All rats underwent right kidney excision and left kidney dissociation, and abdominal walls of rats in sham operation group were sewed up directly ; while the other two groups underwent renal IRI experiment, and curcumin was given before test in curcumin group. Serum creatinine (Cr) and blood urea nitrogen (BUN) of the three groups were determined, renal histopathological changes were observed under light microscope, and TLR4 protein expression was detected by Western blotting. All results were compared. Results Cr and BUN levels in ischemia-reperfusion and curcumin groups were significantly higher than those in sham opration group ( P 〈 0. 05 ) ; Cr and BUN levels in curcumin group were significantly lower than those in ischemia-reperfusion group(P 〈0. 05). There was no obvious change in histopathological damage of renal tubular epithelial cell in curcumin group, while in ischemia-reperfusion group, the cells aggravated obviously. A few of TLR4 expression could be found in rats'nephridial tissue in sham operation group, and TLR4 expressions in ischemia-reperfusion and curcumin groups were significantly higher than that in sham opration group (P 〈 0. 05) ; and TLR4 expression in eureumin group was significantly lower than that in ischemia-reperfusion group (P 〈 0. 05). Conclusion Curcumin may ameliorate renal IRI by inhibition of TLR4 expression.
