人生长激素诱导体外培养的大肠癌细胞株细胞周期动力学变化 被引量:7

EFFECTS OF HUMAN GROWTH HORMONE ON CELL CYCLE KINETICS IN LOVO AND LS-174-T CELLS

摘要 研究人生长激素 (hGH)对大肠癌细胞增殖和分化的影响 .收集人大肠癌细胞株Lovo和LS 174 T(简称 174 ) ,以顺铂和 /或不同浓度的hGH体外培养细胞 ,2 4h后以流式细胞仪测定细胞增殖周期等指标 .结果显示hGH能诱导 174细胞S期数率非常显著地增高 (P <0 .0 1) ;Lovo细胞 10 0ng/mL组Go G1期参数下降 (P <0 .0 5) ,S期参数增加 (P <0 .0 1) ,2 0 0ng/mL组G2 M期百分数增高 .同时用顺铂和hGH处理后 ,两株细胞S期数率均显著下降或无明显上升 ,并显著抑制了Lovo细胞的增殖指数 (P <0 .0 5) ,提高了对 174细胞的DNA抑制率 (P <0 .0 1) ,并且导致 174细胞大量凋亡和死亡 .提示hGH在体外应用于人大肠癌细胞能够使处于DNA合成期的细胞增加 ,为提高肿瘤对周期时相特异性化疗药物的敏感性提供了基础 ,顺铂能够抑制hGH促DNA合成和细胞增殖的作用 。 The effects of human growth hormone (hGH) on cell cycle kinetics were studied in 2 human colorectal cancer cell lines:Lovo and LS 174 T cells. The cells were cultured in RPMI 1640 for 24 hours with or without cisplatin, and with hGH at concentration of 50,100 and 200 ng/mL, respectirely. Cell cycle kinetics was analyzed by flow cytometry. Significant effects of hGH on cell cycle kinetics were found in both cell lines.In LS 174 T, its percentage S phase increased ( P <0.01). In Lovo cells, a decreased percentage of G o G 1 phase ( P <0.01) and increased percentage of S phase ( P <0.01) were found at tha hGH concentration of 100?ng/mL, while the percentage of G 2 M phase elevated at the concentration of 200?ng/mL. hGH and cisplatin inhibited S phase and PI in Lovo cells ( P <0.05), and induced apoptosis of LS 174 T cells. It suggests that hGH may induce proliferation and differentiation of Lovo and LS 174 T cells. Cisplatin may suppress the stimulatory effects of hGH on S phase, and induce apoptosis of cancer cells.
