摘要
大肠杆菌基因敲除技术发展迅速,各种新型技术的出现使得其基因组的改造较以往更为快速、简单,尤其是Red同源重组技术在大肠杆菌基因敲除中的应用已经越来越成熟。综述了几种不同的Red同源重组技术,并分析了每种方法的原理及操作策略,指出了各种方法的特点及优势。
Gene knockout technique of Escherichia coli is developing rapidly. Emerging of various kinds of new technologies has simplified the genome modification of E. coli than before. Especially,the Red homologous recombination system becomes more and more mature in gene knockout of E. coli. In this article, a few different kinds of Red homologous recombination technologies were reviewed. The mechanism and application strategies of each method were analyzed with its characteristics and superiority highlighted.
