摘要
目的比较3种制法的四逆汤抗大鼠急性缺血性心肌梗死模型的药效,确定较佳的制剂方案。方法将Wistar大鼠随机心肌缺血组、假手术组、传统四逆汤组、药典四逆汤组、单药混合四逆汤组。各给药组均预先给大鼠灌胃4 d(2 g/mL),第4日给药30 min后进行急性缺血性心肌梗死手术,6 h后处死动物取血,检测各组血浆CK活性、MDA和SOD含量;用TTC染色测定各组心脏梗死面积。结果与假手术组比较,心肌缺血组血浆CK、MDA含量明显升高;与心肌缺血组比较,传统四逆汤组、药典四逆汤组均能明显降低CK含量;与假手术组比较,心肌缺血组SOD有降低趋势,药典四逆汤组、单药混合四逆汤组有升高SOD趋势;与心肌缺血组比较,药典四逆汤组、单药混合四逆汤组有降低MDA含量的趋势。与假手术组比较,心肌缺血组大鼠心脏梗死率明显增大;与心肌缺血组比较,药典四逆汤组、单药混合四逆汤组心脏梗死率均显著缩小。结论综合分析后认为药典四逆汤组、单药混合四逆汤是治疗大鼠急性缺血性心肌梗死的较佳方案。
Objective: To compared the pharmacodynamics that Sini Decoction with three methods of extraction to anti-acute ischemic myocardial infarction and to determine a better solution for the treatment of anti-acute ischemic myocardial infarction.Methods: Wistar rats were divided randomly into the myocardial ischemia group,the sham-operation control group,the traditional Sini Decoction(SND) group,the pharmacopoeias SND group and THE single-agent mixed SND group.The myocardial ischemia rat group were established by ligation of the left anterior descending coronary artery under 2 mm from starting.The sham-operation group rats were threaded but not ligation.The traditional SND group,the pharmacopoeias SND group,and the single-agent mixed SND group rats were all given SND for 4 days by intragastric administration.The acute ischemic myocardial infarction operation was executed after received SND thirty minutes in the fourth day.These animals were sacrificed after 6 hours and the blood was collected.Then detected the CK activity,the contents of MDA and SOD in blood plasma of every group.TTC stained myocardial infarct size was measured.Results: Comparing with the sham-operation group,the contents of CK,MDA in myocardial ischemia group were significantly increased.Compared with the myocardial ischemia group,the traditional SND group,pharmacopoeias SND group could reduce the contents of CK obviously.Comparing with the sham-operation group,the contents of SOD in myocardial ischemia group had the trend to decrease,and pharmacopoeias SND group,and single-agent mixed SND group had the trend to increase.Compared with the myocardial ischemia group,the contents of MDA in pharmacopoeias SND group,and single-agent mixed SND group had the trend to decrease.Comparing with sham-operation group,myocardial infarct size of the myocardial ischemia group was enlarged remarkably.Compared with the myocardial ischemia group,myocardial infarct sizes of pharmacopoeias SND group and single-agent mixed SND group were significantly reduced.Conclusion: It is showed that SND can remove effectively oxygen free radicals.It can reduce the free radical damage factor,the lipid peroxidation and the release of myocardial enzymes.Meanwhile,It can improve the perfusion of myocardial ischemia and protect positively to myocardial ischemia of the rat.It is considered that pharmacopoeias SND and single-agent mixed SND are the better solutions for the treatment of acute ischemic myocardial infarction by comprehensive analysis.
