复合酶提高黄柏水提效率的关键技术研究 被引量:1

摘要 目的探讨提高黄柏水提工艺效率的方法。方法在黄柏水提工艺程序中,加入纤维素酶-果胶酶-半纤维素酶组成的复合酶,按均匀试验设计考察5因素10水平,用SAS8.2统计分析,测定过滤速度及水提液的浸出物变化作指标,考察酶解工艺条件。结果每克黄柏取相当于酶活力1万U的纤维素酶,0.5万U的果胶酶,0.1万U的半纤维素酶,调温度37℃,pH为3.9的酶解条件下得到的黄柏水提液,分离速度提高5.64倍,黄柏碱与小檗碱收率分别提高2.17、1.14倍。结论应用复合酶可提高黄柏水提工艺的生产效率。 Objective: To study and improve the efficiency of the Cortex Phellodendri Chinensis water-extracting process.Methods: The process of Cortex Phellodendri Chinensis water-extraction was added in composite enzyme which comprises of "cellulose enzyme"-"pectinase"-"half of cellulose enzyme",with the basis on the uniform experimental design investigating the 5 factors and 10 levels,with the SAS 8.2 statistical analysis.The filtration speed and water extraction fluid extract change index was examined enzyme solution process conditions.Results: Each 1g cortex phellodendri taking the enzyme activity was equivalent to 10000 U the cellulose enzyme,5000 U pectinase,1000 U half cellulose enzyme.The temperature was 37 ℃,pH 3.9 of enzyme solution obtained under such condition.The separating speed was increased by 5.64 times,cortex phellodendri alkali and berberine were increased yield 2.17,1.14 times.Conclusion: Application of composite enzyme can improve the Cortex Phellodendri Chinensis water-extracting process production efficiency.
