姜黄素诱导人永生化表皮HaCaT细胞凋亡中Nucleophosmin的表达和定位变化

Expression and Localization of Nucleophosmin During Curcumin-induced Apoptosis in the Immortalized Human Epithelial HaCaT Cells

摘要 该文以姜黄素诱导处理前后的人永生化表皮HaCaT细胞为研究对象,对核仁磷酸蛋白(nucleophosmin,NPM)在核基质中存在、分布及其与凋亡相关基因产物在姜黄素处理前后HaCaT细胞中的共定位关系进行观察研究。Western blot结果显示,NPM存在于人永生化表皮HaCaT细胞核基质蛋白组分中,并在姜黄素处理后的细胞核基质中表达下调,免疫荧光显微镜观察显示,NPM定位在核基质上,经姜黄素处理后出现分布位置与表达水平的变化,激光共聚焦显微镜观察可见NPM与HaCaT细胞中凋亡相关基因Bax、Bcl-2、mtP53和Rb基因产物均存在共定位关系,但在姜黄素处理后细胞中其共定位分布区域出现变化。研究结果证实NPM是一种核基质蛋白,定位于核基质上,NPM在HaCaT细胞诱导凋亡过程中的表达分布及其与凋亡相关基因产物的共定位现象值得进一步探索和研究。 To explore the existence and distribution of nucleophosmin (NPM) in the nuclear matrix and its colocalization with the other apoptosis-related gene products following curcumin treatment in the human epithelial HaCaT cells, the nuclear matrix of HaCaT cells was extracted pre/post curcumin induced apoptosis. Western blot analysis showed that NPM existed in the fractions of nuclear matrix proteins and was down-regulated after curcumin treatment. The immunofluorescence observation revealed that NPM located in the nuclear matrix, curcumin treatment altered its expression level and distribution profile. The colocalization of NPM with the products of apoptosis-related repression genes, including Bax, Bcl-2, mtP53 and Rb, using laser scanning confocal microscopy, were evaluated, and substantial differences were observed following curcumin treatment. The results implied that NPM is a nuclear matrix protein, and the level of its expression and the colocalization with apoptosis-related gene products may play an important role during the apoptosis of HaCaT cells.
