重组人生长激素联合化疗对Bel-7402肝癌细胞的体外影响 被引量:3

Effects of Recombinant Human Growth Hormone Combined with Chemotherapy on the Growth of Bel-7402 Human Hepatic Carcinoma Cell Lines in vitro

摘要 探讨重组人生长激素(recombinant human growth hormone,rhGH)联合化疗药物阿霉素(adriamycin,ADM)对体外培养的Bel-7402肝癌细胞的影响。该实验设计分组为对照组、不同浓度rhGH组、不同浓度rhGH+阿霉素(ADM)组及ADM组,共8组,运用体外细胞培养技术、四甲基偶氮唑蓝比色(methyl thiazolyl terazolium,MTT)和流式细胞技术等研究方法,检测不同浓度的rhGH及不同浓度rhGH+ADM对体外培养的Bel-7402肝癌细胞的生长率、生长曲线、细胞周期及增殖指数(proliferation index,PI)等的影响。48 h后,MTT法检测结果提示:与对照组比较,中、高浓度rhGH组生长率明显升高(P<0.05);ADM组明显抑制Bel-7402肝癌细胞生长(P<0.05);与ADM组比较,rhGH+ADM各组抑制率明显降低(P<0.05)。依据生长曲线图可以得出,rhGH各组较对照组明显升高,尤其rhGH2组升高明显,rhGH各组间变化不明显;rhGH+ADM各组间变化不明显;与rhGH+ADM各组比较,ADM组明显降低。细胞周期结果显示,与对照组比较,rhGH2、rhGH3组的G2/M期比例和增殖指数(PI)显著升高(P<0.05),ADM组的G2/M期比例和PI显著降低(P<0.05),与ADM组相比较,不同浓度rhGH+ADM组的G2/M期比例和PI明显升高(P<0.05)。生长激素可促进体外培养的Bel-7402肝癌细胞生长;生长激素联合阿霉素治疗,可降低化疗药物的抗癌效果。 This study aimed at investigating the effect in hepatocarcinoma Bel-7402 cells by combining recombinant human growth hormone with ADM. Eight groups were assigned as control group, rhGH groups, rhGH+ADM groups and ADM group. Bel-7402 cells were cultured with various concentrations of rhGH and rhGH+ADM. Cell culture technique, MTT assay and flow cytometry were used to assess the growth rate, growth curve, cell cycle and PI on liver tumor cell Bel-7402. The results showed that the growth rate in medium and high concentration of rhGH groups were significantly increased compared with the controls (P〈0.05); ADMgroup obviously decrease the growth of Bel-7402 cells (P〈0.05); compared with the ADM group, inhibition rate of rhGH+ADM groups were significantly decreased (P〈0.05); growth curve indicated that the growth of rhGH groups were increased compared with the control group, especially rhGH2 group; compared with rhGH+ADM groups, growth of ADM group was decreased (P〈0.05). The cell cycle assay indicated that the G2/M phase and PI in the rhGH2 and rhGH3 groups were obviously increased compared with the control group; G2/M phase and PI in rhGH+ADM groups were increased compared with ADM group (P〈0.05). The results suggested that the rhGH stimulated the growth of Bel-7402 hepatic carcinoma cell line in vitro; rhGH combined with ADM will weaken the inhibitory effect of the ADM.
