桦褐孔菌醇对人卵巢癌SKOV3细胞增殖和凋亡的影响 被引量:12

摘要 目的:观察桦褐孔菌醇对人卵巢癌SKOV3细胞株的抗增殖和诱导凋亡作用。方法:选用人卵巢癌SKOV3细胞株进行体外培养,分为不同浓度桦褐孔菌醇组(25μg/ml、50μg/ml、100μg/ml)和对照组(未加桦褐孔菌醇)。分别采用四甲基偶氮唑蓝(MTT)比色法测定两组对人卵巢癌SKOV3细胞株的作用;应用倒置显微镜、HE染色光学显微镜观察两组细胞形态学变化;通过免疫组化法比较两组核增殖抗原Ki-67的表达。结果:①作用48小时后,桦褐孔菌醇浓度为25μg/ml、50μg/ml、100μg/ml的抑制率分别为(17.16±4.71)%、(35.16±4.26)%和(57.83±3.48)%,与对照组(0)比较差异有统计学意义(P<0.05)。②HE染色光镜下,桦褐孔菌醇组卵巢癌SKOV3肿瘤细胞体积变小、变圆,核染色质浓缩或染色质块形成,有的细胞膜起泡形成凋亡小体;倒置显微镜下,桦褐孔菌醇组SKOV3肿瘤细胞体积变小、变圆,核染色质凝集,细胞间连接疏松,贴壁能力明显减弱。③作用48小时后,桦褐孔菌醇浓度为25μg/ml、50μg/ml、100μg/ml的Ki-67阳性表达率分别为(64.67±3.98)%、(43.83±3.06)%和(34.00±2.61)%,与对照组(92.83±3.06)%比较,差异均有统计学意义(P<0.05)。结论:桦褐孔菌醇对人卵巢癌SK-OV3细胞株有抗增殖作用和诱导凋亡作用,并表现出浓度依耐性。 Objective:To explore the effect of inotodiol on cell proliferation and apoptosis in human ovary cancer SKOV3 cells. Methods:SKOV3 human ovary carcinoma cells were cultured in vitro,and divided into different concentrations of inotodiol group (25,50,100 μg/ml) and control group (without inotodiol). MTT assay was used to determine the effect of cell growth inhibitory in two groups. Morphological changes of SKOV3 cells in two groups were observed through invert microscope and by HE staining under light microscope. Expressions of Ki-67 were detected by immunocellulerchemical method. Results.①SKOV3 cells were treated with inotodiol for 48 hours,inotodiol could inhibit the proliferation of SKOV3 human ovary carcinoma cells in dose-dependent manner, inhibition rates of SKOV3 cells treated with inotodiol in concentrations of 25 μg/ml, 50 μg/ml,100 μg/ml were (17.16 ± 4. 71 )%, (35.16 ± 4.26)% and (57.83 ± 3.48)% respectively, with significant difference compared with the control group ( P 〈 0. 05). GAffer HE staining, the SKOV3 human ovary carcinoma cells treated with inotodiol showed morphological changes such as cellular volume small and round, nuclear chromatin condensation,plasma membrane bleb formation,their apoptosis body formations were also observed under light microscope. The SKOV3 human ovary carcinoma cells treated with inotodiol showed cellular volume small and round, nuclear chromatin condensation, loose between the cells and adherent ability obviously decrease under invert microscope. ②The positive expression rates of Ki-67 in SKOV3 cells after treated with inotodiol for 48 hours in concentrations of 25 μg/ml,50 μg/ml, 100 μg/ml were (64. 67 ±3. 98)%, (43. 83 ±3.06)% and (34. 00 ±2.61 )%, respectively,there was significant difference compared to the control group (92.83 ± 3.06) % ( P 〈 0. 05). Conclusions: Inotodiol has the effect of an- ti-proliferation and apoptosis inducing in human ovarian cancer cell line SKOV3, which is in a concentration dependent maner.
