ADSCs复合纤维蛋白凝胶修复大鼠坐骨神经缺损

ADSCs combined with fibrin gel in repairing sciatic nerve defect in rat experimental study

摘要 目的研究脂肪间质干细胞(ADSCs)复合纤维蛋白凝胶对大鼠坐骨神经缺损修复的效果。方法体外分离、培养、鉴定sD大鼠ADSCs。建立大鼠坐骨神经缺损模型。在神经缺损处分别植入:A组,实验组(ADSC+纤维蛋白凝胶+脱细胞神经导管);B组,纤维蛋白凝胶组(纤维蛋白凝胶+脱细胞神经导管);C组,生理盐水组(生理盐水+脱细胞神经导管);D组,自体坐骨神经移植组。术后进行大体观察、坐骨神经功能指数测定、腓肠肌湿质量恢复率测定和组织学观察。结果A组和B组均可见再生神经,直径较D组神经干细,C组无再生神经组织生成。术后第8、12周:A组坐骨神经指数恢复情况优于B组,差异有统计学意义(P〈0.01);D组坐骨神经指数恢复情况优于A组,但差异无统计学意义(P〉0.05)。A、B、D组腓肠肌湿质量恢复率明显高于C组,差异有统计学意义(P〈0.01),A组与B组比较差异无统计学意义(P〉0.05)。结论在纤维蛋白凝胶中加入ADSCs并将其植入脱细胞神经导管内,再生神经显示出更好的结构和功能,结果表明在神经再生时加入ADSCs能够促进周围神经的再生。 Objective To study the adipose-derived mesenchymal stem cells (ADSCs) combined with fibrin gel on rat sciatic nerve defect repair effect. Methods Isolation, culture, and identification of SD rats ADSCs was performed in vitro. Rat sciatic nerve defect model was established. The nerve defects were implanted into experimental group (Group A, ADSCs + fibrin gel + acellular nerve conduit), fibrin gelatin group (Group B, fibrin gel + acellular nerve conduit) , the physiological saline group (Group C, saline + acellular nerve conduit) , and autologous sciatic nerve transplantation group (Group D). Postoperative gross observation was performed on sciatic nerve function index, gastrocnemius muscle wet weight recovery rate test and histological observation. Results Group A and B showed the nerve regeneration, the larger diameter autologous sciatic nerve transplantation group neural stem cells, Group C had no regeneration of nerve tissue. After 8 and 12 weeks, the sciatic index recovery of Group A was better than that of Group B, and the difference was statistically significant ( P〈0.01 ) ; The sciatic index recovery of Group D was better than that of Group A, but the difference was not statistically significant ( P 〉 0.05 ). The gastrocnemius muscle wet weight recovery rates in Group A, B and C were significantly higher than those in Group C, and the difference was statistically significant (P 〈 0. 01 ), while there was no statistical significance between Group A and D (P 〉 0. 05 ). Conclusion Infibrin gel with ADSCs into acellular nerve conduit, the regenerating nerve showed a better structure and function, and the results show that in nerve regeneration chamber into ADSCs can promote the re- generation of peripheral nerve.
