摘要
以栀子苷粗提取物为原料,采用高速逆流色谱法分离栀子苷,溶剂系统为A:乙酸乙酯∶正丁醇∶水(2∶1.5∶3)和B:正丁醇∶水(1∶1),上相为固定相,下相为流动相,流速为2.0 mL/min,转速为850 r/min,温度控制在25℃,纯度用HPLC测定。结果表明,利用溶剂系统A和B进行HSCCC制备栀子苷,使栀子苷含量从50.75%(HPLC)分别提高至86.6%和91.8%,回收率分别为81.36%和78.12%。
Geniposide was isolated and purified by high speed counter counter current chromatography (HSCCC) in semi -preparative scale from the crud extract of the fruits of Gardeniajusminoides El. Separation was performed with solvent system A: ethyl acetate: n-butanol: water (2: 1.5: 3) and B: n-butanol : water( 1 : 1 ) by eluting the lower aqueous phase at a flow rate of 2.0 mL/min and a revolution speed of 850 r/min, at 25~C. After the proceeding of separation in solvent system A and B, 160.0 mg( HPLC are- as giving a purity of 86.6% ,the rate of recovery of HSCCC is 81.36% ) and 205.0 mg(a purity of 91.8%, the rate of recovery of HSCCC is 78.12% ) geniposide were obtained.
