摘要
用噻唑蓝 (MTT)比色分析法检测了在正常及去肾上腺大鼠双侧海马内微量注射去甲肾上腺素 (Noradrenline,NA)对刀蛋白 (ConA)刺激的脾淋巴细胞增加增殖反应的影响 结果 :(1 )在正常大鼠 ,NA(4μl,6 0× 1 0 -3 mol/L)、β1 受体激动剂Dobutamine(Dob,4μl,6 0× 1 0 -3 mol/L)和 β2 受体激动剂Metaproternol(Met,4μl,8 0× 1 0 -3 mol/L)均可抑制ConA刺激的脾淋巴细胞增殖反应 其中NA的作用最强 ,Met次之 ,Dob的作用最弱 ;α及 β受体阻断剂酚妥拉明 (Phen ,2 μl,2 0× 1 0 -3 mol/L)和心得安 (Prop,2 μl,2 0mol/L)均可部分阻断NA的免疫抑制作用 ,且Prop的作用更强 (2 )在去肾上腺组 ,NA的免疫抑制作用不明显 提示 :海马内NA对机体的细胞免疫功能具有明显的抑制作用 ,此作用是由α及β受体共同介导 ,其中 ,β受体的作用大于α受体 ,β2 受体的作用大于 β1 受体 此外 。
In the present work, the effect of intrahippocampl microinjection of noradrenaline(NA) and its receptors antagonists and agonists on cellular immune functions were investigated in normal and adrenalectomy rat by determing the proaliferative activity of Con A_stimulated splenic lymphocytes in MTT mothed, the results showed: (1) In normal group, the prolifertative activity of ConA_Stimulated spenic lymphocytes were inhibited with microinjection NA and β 1_,β 2_adrenergic receptors agonists Dobutamine( Dob, 4μl, 6 0×10 -3 mol/L), Metaproternol(Met, 4μl, 8 0×10 -3 mol/L), compared with their intensity of effect, NA>Met>Dob; the immunosuppression effect induced by NA was partly hindered by α_and β_receptor antagonists, phentolamine(Phen, 4μl, 8 0×10 -3 mol/L) and propranolon( Prop, 4μl, 8 0×10 -3 mol/L), and the action of Prop was more evident (2)In adrenalectomy group, immunosuppression effect induced by NA was inconspicuousness, the results suggested that NA inhippocampus can inhibit distinctly cellular immune functions, which was predominantly mediated by β 2 adrenergic receptor with a minor contribution of β 1_an a_adrenergic receptors Moreover, keeping intact construction and function of adrenal gland have an important role in the efflect of NA on cellular immune function
