摘要
[目的]建立使用黄曲霉毒素B1单克隆抗体修饰金电极测定饲料中黄曲霉毒素B1的新方法。[方法]试验采用电化学方法优化检测条件,建立标准曲线并对样品进行检测。[结果]试验得出,黄曲霉毒素B1单克隆抗体修饰金电极检测饲料中黄曲霉毒素B1的最优条件为:电极修饰条件为4℃、8 h,磷酸盐缓冲溶液的pH为7.4。该检测方法对黄曲霉毒素B1的线性检测范围为1.427~150.000ng/ml,加标回收率在94.44%~101.36%。[结论]使用该免疫电化学方法,检出限较低,可以用于饲料中黄曲霉毒素的检测。
[Objective] A new method for determining the AFB1 in feed was established based on aAFB1 Modified Au Electrode.[Method] The optimal conditions,calibration curve and determination of Aflatoxin B1 in feed all were got by electrochemical methods.[Result] The optimal conditions: the Au electrode should be modified by 4 ℃,8 h,the optimal value of PBS is 7.4.Under the optimized conditions,the linear range is 1.427-150.000 ng/ml,the recovery rate is from 94.44% to 101.36%.[Conclusion] The immunoelectrochemical method has a lower detection and can be applied to determination of AFB1 in feed successfully.
