麦糟固态发酵生产纤维素酶 被引量:7

摘要 以麦糟为主要原料 ,利用里氏木霉 (TrichodermareeseiRutC 30 )固态发酵生产纤维素酶 ,在浅盘发酵中 ,通过对发酶过程品温和曲料pH的优化控制 ,得到了优化后的工艺条件。在发酵前期品温控制在 2 7~ 30℃ ,后期最高不超过 35℃ ,最高酶活达到 611U/g曲。当曲料pH第一次回升至 4 .3~ 4 .5时 ,用稀盐酸进行喷酸控制。采用优化后的pH和品温控制工艺 ,FPA酶活达到 50 8U/g曲 ,相当于 2 12 Cellulase production was carried out by solid state process with Trichoderma reesei Rut C 30 when distiller′s wheat was employed.In the trays,the optimized koji temperature was as follows:before 48?h koji temperature should be controlled between 27~30?℃,after 48?h below 35?℃,and maximal FPA activity of 611?U/g was achieved.When pH value increased back to 6.0~6.2,FPA activity ceased to increase or declined.Based on the investigated data pH control strategy was proposed to be carried out when pH rised back to 4.3~4.5 for the first time.Spraying of inorganic acid into substrate,such as HCl,could counteract the increase of pH during cultivation.With pH control technology,43.7?% improvement of FPA activity was estimated and the results clearly illustrated that pH control enhanced cellulase production.With koji temperature and pH control technology,FPA activity of 508?U/g koji(about 2120 U/g cellulose) was obtained.The pH control technology could be carried out in large scale solid state fermentation of cellulase.
