摘要
目的 为进一步研究白念珠菌保护性抗体免疫以及系统性念珠菌病早期诊断奠定基础。方法 运用细胞融合、间接ELISA、IIF等技术,制备抗白念珠菌胞壁外膜单克隆抗体。结果 建立了三株分泌抗白念珠菌胞壁外膜成分单克隆抗体杂交瘤细胞株-IB5、3E8、4C7;单抗亚类分析显示1B5为IgM,3E8、4C7为IgG1;这三株单抗不仅能使白念珠菌孢子表面染上荧光,也能使芽管表面带上荧光。结论 制备成功三株抗白念珠菌胞壁外膜成分单克隆抗体。
Objective:To provide a basis for further studying the protective immunity to Candida albicans with antibody and early diagnosis of Candida albicans Methods:Wonoclonal antibody (Mab) against outer membrane of cell - wall of Candida albicans was prepared by cell fusion, in- direct Elisa and IIF technic. Results:Three strains of hybridoma, IB5, 3E8 and 4C7 were established. Analyzing Mab immunoglobulin isotype sub- class, we found that 1B5 belonged to IgM, 3E8 and 4C7 belonged to IgG. By indirect immunofluorescence assay, all of the three MAbs could stain not only the surface of yeast cell wall but also the surface of germ tuber of Candida albcians. Conclusion:three monoclonal antibodies against outer membrane of cell wall of Candida albicans were successfully prepared.
