摘要
目的 探讨比较阿霉素 (ADM)化疗与 43℃加热化疗对人肝癌细胞 - 772 1(以下简称 772 1细胞 )和耐药人肝癌细胞模型 - 772 1/Adm(以下简称 772 1/Adm细胞 )的细胞毒作用 .方法 以体外培养的人肝癌细胞 - 772 1和我们前面培养建立的人肝癌细胞模型 - 772 1/Adm为研究对象 ,采用水浴加温法 ,体外细胞毒试验 (MTT法 ) ,观察阿霉素 (ADM)化疗与加热化疗对细胞的生长抑制规律的影响 ;采用流式细胞技术检测加热对 772 1细胞和 772 1/Adm细胞胞内阿霉素浓度的影响 .结果 1772 1/Adm细胞对阿霉素的敏感性明显低于772 1细胞 ;2加热可以明显提高两种细胞对阿霉素的敏感性 ;3流式细胞检测显示加热可明显提高这两种细胞 ,尤其是772 1/Adm细胞内的阿霉素浓度 .结论 加热可以显著提高人肝癌细胞 - 772 1和耐药人肝癌细胞模型 - 772 1/Adm对阿霉素的敏感性 .
AIM To comparatively study the cytotoxic effects of chemotherapy and thermochemotherapy of ADM on human hepatocellular carcinoma cell line (7721) and human multidrug resistant hepatocellular carcinoma cell line ( 7721/Adm). METHODS By means of water bath and MTT assay, the inhibition pattern of cell growth was observed. The effects of hyperthermia on intracellular concentration of ADM of 7721 and that of 7721/Adm were detected by flow cytometry. RESULTS ① The sensitivity of 7721/Adm to ADM was significantly lower than that of 7721;②Hyperthermia remarkably increased the sensitivity of 7721 and 7721/Adm to AMD; ③Hyperthermia increased the intracellular concentration of ADM in those two sorts of cells mentioned above, especially in 7721/Adm. CONCLUSION Hyperthermia can increase the sensitivity of 7721 and 7721/Adm to ADM.
