Effects of 2,4-D and NAA on Embryogenic Callus Formation and Shoot Regeneration of Sugercane (Saccharum spp.)

摘要 以新台糖22(ROC22)、福农94-0403、云蔗03-258及P44为材料,MS为基本培养基,研究切割厚薄不同的外植体及2,4-D浓度对甘蔗愈伤组织诱导的影响,R1、R2及R3三种不同培养基对分化不定芽的影响。结果表明:①外植体切片0.1cm时的诱导率较切段0.5cm高;②切片时最适2,4-D浓度为1~2mg/L,而切段时则以3mg/L时诱导率达最大;③R2和R3分化效果优于R1,R3分化培养基:MS+6-BA 1mg/L+NAA 1mg/L对愈伤组织分化不定芽的效果最佳。 Abstract: Explants of sugarcane varieties ROC22, Funong 94-0403, Yunzhe 03-358 and P44 were cultured on MS medium with various supplements. Experiments were executed to evaluate the effect of different thickness of explants and different concentration of 2,4-D on callus induction as well as three different medium of R1, R2, R3 on shoot regeneration. The results were as the following: firstly, the induction rate of explants of 0.1 cm thick discs was higher than that of 0.5 cm thick segments; secondly, the appropriate 2,4-D concentration for callus induction were 1-2 mg/L for explants of 0.1 cm thick and 3 mg/L for explants of 0.5 cm thick; thirdly,shoot regeneration rating Of the R2 and R3 were better than that of the R1, and the R3 medium: MS supplemented with NAA at 1 mg/L and 6-BA at 1 mg/L was the best medium for shoot regeneration.
