大黄素抑制SHG44胶质瘤细胞增殖及诱导凋亡的研究

摘要 目的研究大黄素对人胶质瘤SHG44细胞增殖抑制及诱导凋亡的作用。方法实验组大黄素浓度分别为10,20,40,80μmol/L,并设对照组(未经药物处理),共5组。用噻唑蓝法(MTT)观察大黄素对SHG44细胞的增殖抑制作用;用流式细胞仪法分析大黄素的诱导凋亡作用,通过比色法测定细胞内caspase-3的相对活性。结果与对照组相比,大黄素能明显抑制SHG44细胞的生长,且具有浓度、时间依赖性,其IC_(50)(48 h)为24.79μmoL/L;大黄素处理胶质瘤SHG44细胞后,流式细胞仪分析大黄素能浓度依赖性诱导SHG44细胞凋亡;大黄素作用SHG44细胞后,处理组caspase-3的相对活性明显升高,并存在时间依赖关系。结论大黄素能抑制胶质瘤SHG44细胞的增殖并诱导细胞凋亡,其机制可能与caspase-3活化有关。 Objective To explore the effects of emodin on proliferation and apoptosis of human glioma cells SHG44.Methods The cultured SHG44 cells were treated with 10,20,40 and 80μmol/L emodin in experimental groups,respectively,but not treated in control group.The inhibitive effect of emodin on SHG44 cell proliferation was detected by MTT assay,cell apoptosis was detected by flow cytometry, and the relative activity of caspase-3 was detected by colorimetry.Results Compared with control group,emodin significantly inhibited the proliferation of SHG44 cells in a dose-dependent and time-dependent manner,and the CI_(50)(48 h) of emodin was 24.79μmol/L.Flow cytometry analysis showed that emodin dose-dependently induced the apoptosis in SHG44 cells after 12 h treatment(P 0.05).The relative activity of caspase-3 was elevated time-dependently after emodin treatment(P 0.05).Conclusion Emodin can inhibit the proliferation of glioma cells SHG44 and induce the apoptosis in SHG44 cells,which may be related to the caspase-3 activation.
