摘要
目的观察不同剂量、不同作用时间芹菜素对雄性小鼠肝脏总抗氧化能力(T-AOC)、还原型谷胱甘肽/氧化型谷胱甘肽(GSH/GSSG)、丙二醛(MDA)含量的影响。方法 200只成年SPF级昆明种雄性小鼠随机分为对照组(生理盐水10 mL/kg)、低剂量组(芹菜素252 mg/kg)、中剂量组(芹菜素504 mg/kg)和高剂量组(芹菜素1008 mg/kg)。连续灌胃7、14、21、28、35 d,分批处死,称取肝脏制备匀浆,测定T-AOC、GSH、GSSG及MDA含量。结果高剂量组T-AOC含量[(1.397±0.641)U/mg]显著低于对照组[(1.573±0.781)U/mg](P=0.000);芹菜素灌胃28 d GSH/GSSG值相对对照组升高幅度最大(140.556%);高剂量组灌胃21 d MDA含量[(1.068±0.729)nmol/mg]显著高于对照组[(0.354±0.212)nmol/mg](P=0.000)。结论 1008 mg/kg芹菜素对小鼠肝脏组织T-AOC具有一定的抑制作用,并可增加肝脏脂质过氧化程度。T-AOC可以比较早地反映出机体抗氧化能力的变化。
Objective To observe the effects of apigenin(AP) at different doses and time on total antioxidant capacity(T-AOC),reduced glutathione(GSH)/oxidized glutathione(GSSG),maiondialdehyde(MDA) in livers of male mice.Methods 200 SPF Kunming mice were randomly divided into the control group(saline 10 mL/kg),low-dose group(AP 252 mg/kg),middle-dose group(AP 504 mg/kg) and high-dose group(AP 1008 mg/kg).The medicines or saline was given by intragastric administration.In batches,10 mice of each group were killed and then prepared liver homogenate at 7,14,21,28 and 35 days after treatment.Supernatant were collected from the homogenate by centrifugation,then T-AOC,MDA,GSH and GSSG were measured,respectively.Results T-AOC of the high-dose group [(1.397±0.641) U/mg] was significantly lower than that of the control group [(1.573 ±0.781) U/mg](P=0.000).Compared with the control group,GSH/GSSG increased the most after treated by AP for 28 days(140.556%).MDA content of high-dose group [(1.068 ± 0.729) nmol/mg] was higher than that of the control group [(0.354±0.212) nmol/mg] at 21 d after treatment(P=0.000).Conclusion 1008 mg/kg AP can inhibit T-AOC in a certain level,and promote the lipid peroxidation of male mice.The early change of the antioxidant capacity can be shown by the T-AOC in liver.
