摘要
【目的】初步阐明针灸对缺氧大鼠神经保护作用的机制。【方法】健康雄性SD大鼠随机分为缺氧组和针灸治疗组,每组12只。针灸治疗组大鼠每日针灸关元、百会和内关3穴,缺氧暴露0、12、24、36 h后处死取大脑皮层,分别采用RT-PCR方法和免疫印迹Western blotting法检测大脑皮层促红细胞生成素(Erythropoietin,EPO)mRNA和蛋白的表达情况;采用普通酶联免疫吸附实验测定大鼠血清中神经特异性烯醇化酶含量。【结果】缺氧12、24、36 h条件下,针灸治疗组大鼠大脑皮层EPO mRNA和蛋白表达量较同等缺氧暴露条件下对照组表达量提高,针灸治疗组大鼠血清NSE含量较同等缺氧暴露条件下对照组降低,缺氧暴露24、36 h条件下,针灸治疗组大鼠血清中NSE含量比对照组显著降低。【结论】针灸能够促进缺氧大鼠大脑皮层EPO的表达,可能在其保护缺氧脑组织中起着重要作用。
【Objective】To study the effect of acupuncture on erythropoietin(EPO)expression in the Brain Cortex of Hypoxia Rat.【Methods】RT-PCR and Westren Blot were used to test EPO expression,and the concentrations of serum NSE was measured by the enzyme-linked immunoassays(ELISAs) kit.【Results】Acupuncture could significantly enhance the EPO mRNA and protein expression under anoxic environment in the rat brain cortex,and the concentrations of serum NSE was significantly lower in 24 h and 36 h acupuncture group than in the control group(P 0.05).【Conclusion】Enhancing the expression of EPO may play an important role in the neuroprotective effect of acupuncture in the brain cortex of hypoxia rat.
