中药总蒽醌粗提物对低密度脂蛋白氧化的抑制作用

Inhibition of low density lipoprotein oxidation by anthroquinone extracted from Chinese herbal medicine

目的 从单味中药大黄、虎杖、何首乌中提取总蒽醌类化合物 ,观察其对低密度脂蛋白氧化修饰的抑制作用。方法 以95%乙醇提取并加用碱提取酸沉淀法提取蒽醌类成分 ,观察其对 Cu2 + 诱导的 LDL氧化的抑制作用 ,以硫代巴比妥酸反应物质法(TBARS)测定丙二醛 (MDA)含量 ,计算药物对氧化的抑制率以及抑制率为 50 %时所需药物浓度 (IR5 0 )。结果 总蒽醌类提取物使氧化动力曲线迟滞相延长 ,峰值降低 ,抑制过氧化脂质生成并有浓度依赖性 ,各种药物的 IR5 0 分别为 8.1 8μg/ ml(大黄 )、1 5.58μg/ ml(虎杖 )、3.0 6× 1 0 9μg/ ml(何首乌 )。结论 三种药物蒽醌类提取物均有抑制 LDL氧化作用 ,但抑制作用强度不同。

Independently extracted the anthroquinone mixture from herbal medicine Rheum palmatum L, Polygonum cuspidatum Sieb et Zuu and Polygonum multiflorum Thunb were examined.Methods Extract the anthroquinone constituent with 95% ethanolic solution. The extracts solved in alkaline solutions and then deposit from acidic solution. The oxidative low density lipoprotein(ox LDL) was induced by Cu 2+ and oxidation degree was assessed by measuring thiobarbituric acid reactive substances(TBARS). The inhibition rate and IR 50 was assessed by bivariate correlation and linear regression with SPSS.Results The extracts could delay the lag phase in the kinetics process of LDL oxidation. The formation of TBARS was inhibited.IR 50 was 8 18 μg/ml、15 58 μg/ml、3 06×10 9 μg/ml respectively.Conclusions All the extracts can inhibited the LDL oxidation. The antioxidative activity of the three extracts were different, Rheum palmatum L seems to be the most powerful one.
