摘要
为了探讨肝切除后肝细胞生长因子 (HGF)及增殖细胞核抗原 (PCNA)在肝再生中的作用 ,制备大鼠肝切除后肝再生模型 ,应用免疫印迹法及免疫组织化学染色分别检测 HGF的分子形式及 PCNA的表达。结果发现 :HGF在肝切除后残存的肝组织中的含量在 12 h显著增加 ,是正常的 14倍。增加的水平维持到 2 4h。但 HGF的重链带低于检测水平。PCNA在肝切除后残存的肝组织中的表达明显升高 ,于术后 2 4h达高峰。阳性细胞高达 5 4%。提示 HGF在肝再生中的作用是微不足道的 ,而 PCNA可以作为一种肝细胞增生的指标。
The role of hepatocyte growth factor (HGF) and proliferating cell nuclear antigen (PCNA) in liver regeneration after partial hepatectomy was studied. The model was established by partial hepatectomy. The molecular form of HGF and the expression of PCNA was determined by using immunobloting and immunohistochemical methods, respectively. The results showed that amount of HGF after partial hepatectomy was progressively increased at the first 12 h with the maximum level being about 14 fold higher than that of the normal rat liver and maintained up to at least 24 h. The band corresponding to the heavy chain of HGF was lower than the detectable level. The expression of PCNA was increased in remnant liver tissue and reached the highest peak 24 h after partial hepatectomy. The percentage of positive cells was as high as 54 %. It was concluded that HGF might play a slight role in liver regeneration and PCNA could be used as an important index of cell regeneration in liver regeneration after partial hepatectomy.
