摘要 目的研究肝癌转铁蛋白受体 (TfR)的基因表达及其单克隆抗体免疫诊断肝癌的价值。 方法对 2 0例肝癌手术标本包括癌巢组织、癌旁组织及周围正常组织进行Northernblot分析 ,了解三种组织TfRmRNA表达情况 ;通过间接免疫荧光及免疫组织化学ABC法了解肝癌细胞和正常肝细胞膜上TfR表达情况。 结果TfRmRNA表达癌组织最高 ,癌旁组织次之 ,周围正常组织最低 ,后两者间无显著差异 ;肝癌细胞膜转铁蛋白受体荧光阳性率明显高于正常肝细胞 ,两者有显著差异。免疫组织化学ABC法中 1 9例强阳性染色 ,1例阴性。 结论转铁蛋白受体的基因表达在肝癌组织中明显增高 。 Objective To study the transferrin receptor(TfR) gene expression and value of TfR monoclonal antibody in the diagnosis of hepatocellular carcinoma. Methods Twenty specimens of hepatocellular carcinoma taken in operation including the cancer nests tissues, the perineoplasmic tissues, and the normal liver tissues were analyzed by Northern blot, and indirect immunofluorescene and immunoperoxidase were used as well. Results The highest mRNA expression of TfR occurred in the cancer nests tissues, secondly, in the perineoplasma tissues, and the lowest in the normal liver tissues; but the latter two had no significant differences. The positive rate of indirect immunofluorescence of the hepatocellular carcinoma cells was higher than that of the normal liver tissues with significant differences. In the immunoperoxidase ABC method, 19 specimens were intensely positive, but 1 was negative. Conclusion TfR gene expressions increase obviously in the hepatocellualr carcinoma cells. It is suggested that TfR monoclonal antibody may be an ideal carrier of immunodiagnosis to hepatocellular carcinoma.
