乌司他丁对大鼠缺血-再灌注肝脏Toll样受体表达的影响 被引量:1

摘要 目的观察乌司他丁对大鼠肝脏缺血-再灌注损伤以及缺血肝叶Toll样受体2(TLR2)表达的影响。方法SD大鼠36只,随机分为假手术组、缺血-再灌注组(I/R组)及乌司他丁组。于再灌注120min后采集标本,测定血清ALT、AST,取缺血-再灌注肝叶组织,测定组织超氧化物歧化酶(SOD)及丙二醛(MDA)活性,实时定量PCR法测定TLR2表达水平。结果乌司他丁组大鼠血清ALT、AST和肝组织MDA水平低于I/R组(P〈0.05);肝组织S01)水平高于L/R组(P〈0.05);再灌注后TLR2表达水平明显升高,而乌司他丁组TLR2表达低于I/R组(P〈0.05)。结论乌司他丁可以对部分肝叶缺血引起的再灌注损伤有保护作用,其机制可能为抑制TLR2的表达。 Objective To investigate the influence of ulinastatin on liver ischemia - reperfusion injury and the expression of toll - like receptor 2 (TLR2) using liver ischemia - reperfusion rat model. Methods 36 SD rats were divided into sham group, ]/R group and u]inastatin group. Blood sample and liver tissue were collected after t20 min of reperfusion. Serum ALT, AST and tissue SOD, MDA were measured. The expression of TLR2 was measured by real -time PCR. Results The serum ALT, AST and the tissue MDA in ulinastatin group is lower than that of I/R group(P 〈0.05). The tissue SOD is lower in ulinastatin group( P 〈 0.05 ). The expression level of TLR2 is lower in ulinastatin group than I/R group. Conclusion Ulinastatin can protective liver from ischemia - reperfusionon injury. The mechanism of this protective effect may be the suppression of TLR2 expression.
