摘要
目的 :测定我国 3株登革 2型病毒 (dengue 2virus ,DEN2 )流行株的全序列并确定其基因型。方法 :运用RT PCR和 5′ ,3′RACE法测定我国 3株登革病毒的全序列 ,采用邻结法绘制系统发生树。结果 :DEN2 0 4、4 3、4 4株的核苷酸及氨基酸差异分布于整个序列之中 ,DEN2 0 4与DEN2 4 3、4 4共有 2 1个氨基酸位点的差异引起极性或电荷变化。DEN2 0 4株与JAM株、越南分离株和泰国分离株同为Ⅲ基因型 ,DEN 2 4 3、4 4株与台湾、菲律宾和新几内亚株分在Ⅱ基因型。结论 :我国不同地区存在同一基因型的DEN2传播 ,同一地区存在不同基因型的DEN2传播。
Objective: To determine the complete genomic sequences and genotypes of three dengue 2 virus strains isolated in China. Methods: The complete genomic sequences of three dengue 2 virus strains were determined by RT-PCR and RACE. The phylogenetic trees were analyzed by neighbor-joining method. Results: Nucleotide and amino acid changes were random in the genomes of DEN2-04, 43, and 44. Twenty-one amino acid substitutions confered a charge difference or side chain polarity between DEN2-04 and DEN2-43, 44. DEN2-04 strain fell within the group Ⅲ comprising JAM and strains from Thailand and Vietnam, while DEN2-43 and DEN2-44 strains fell within the group Ⅱ comprising NGC and strains from Taiwan and Philippines. Conclusions: One genotype of dengue 2 virus may be distributed in different regions,while there are two or more genotypes in one region in China. [
