摘要
目的 :对 1999年 8月福建省福州市暴发的不明热的病毒病原进行分离与鉴定。方法 :首先采用传代蚊细胞的微量培养方法进行病毒分离 ,并通过脑内接种途径观察对乳小鼠的致病性。然后经间接免疫荧光法和RT PCR技术对毒株进行型别鉴定。结果 :患者恢复期血清标本中存在登革 2型病毒特异抗体。从急性期血清标本中分离出 11株病毒 ,在传代蚊细胞中可产生稳定的细胞病变并对乳小鼠致病。其基因组为登革 2型病毒特异的RNA分子。结论 :福建省福州市流行的不明热疾病是由登革 2型病毒感染所致。
Objective:To isolate and identify the viral pathogen in an outbreak of unknown fever in August 1999 in Fuzhou,Fujian Province. Methods:Firstly, the virus was isolated from acute phase sera of patients by the subcultures of mosquito cell and its pathogenicity in suckling mouse was observed after intracerebral inoculation. Then, serotype of the virus strain was identified by the indirect immunofluoresence and RT-PCR techniques. Results:The specific antibody against dengue virus was detected from convalescent sera of patients. Eleven strains of virus were isolated from the acute phase sera. The isolated virus strains could produce stable cytopathic effect (CPE) in the subcultures of mosquito cell and were pathogenic to suckling mouse. The genome of the isolated virus was RNA and specific to dengue-2 virus. Conclusions:An epidemic of unknown fever disease among many people was caused by infection of dengue virus type 2 in Fuzhou,Fujian Province. [
