摘要
目的 :自行合成LewisY拟糖脂并寻找一种合适的标记拟糖脂的方法。方法 :应用LewisY四糖合成了LewisY拟糖脂 ,以此制备了含3H 胆固醇标记和LewisY拟糖脂的脂质体 ,利用LewisY糖特异性的抗体鉴定了糖脂的免疫学活性 ,并检测了LewisY拟糖脂 脂质体与该抗体的结合能力。结果 :在适宜条件下 ,LewisY四糖拟糖脂的合成率为 4 0 % ,合成的特异性糖脂高效薄层层析为单一条带 ;制备的LewisY拟糖脂 脂质体与Lewis寡糖抗体具有特异结合能力。结论 :以3H 胆固醇标记的LewisY拟糖脂 脂质体是一种效果良好的间接标记的拟糖脂探针。
Objective:To explore convenient labeling method for using neoglycolipid as a probe to investigate the biological function of oligosaccharide. Methods:Neoglycolipid containing Lewis Y was chemically synthesized, then was incorporated into liposome labeled with tritium-cholesterol.The immunoactivity of the Lewis Y glycolipid on HPTLC and binding ability of Lewis Y-liposome to specific antibody AH6 direct to Lewis Y oligosaccharide were examined. Results: Under optimum condition, nearly 40% product of Lewis Y glycolipid,which was single band on HPTLC, was obtained.The tritium labeled liposome containing Lewis Y glycolipid showed the binding ability to AH6 antibody. Conclusions: Lewis Y-liposome labeled with tritium-cholesterol is a useful indirectly labeled neoglycolipid probe. [
