摘要
目的 研究IL 1和IL 2对甲状腺细胞生成甲状腺球蛋白 (Tg)的调节作用。方法 采用Graves病 (GD)甲状腺组织 ,以原代细胞培养及放免分析技术 ,检测两类细胞因子刺激后Tg的分泌状况。结果 基础状态下 ,10 -3 ~ 10 3 U/ml的IL 1和IL 2对甲状腺细胞产生Tg的功能无明显影响 ;IL 1剂量依赖性地抑制促甲状腺激素 (TSH)对甲状腺细胞分泌Tg的刺激效应 ,而IL 2则促进TSH诱导下Tg的产值。结论 IL 1和IL 2是重要的甲状腺生物调节剂。
Objective To evaluate the effect of interleukin-1 (I L-1) and interleukin-2(IL-2) on the generation of thyroglobulin (Tg) from cu ltured thyroid cells. Methods Thyroid tissues obtained from patients with Graves′ disease (GD) were digested with collagenase and trypsin and incub ated with or without recombinant IL-1 and IL-2 in the absence or presence of T SH. Tg in the supernatant was measured by radioimmunoassay. Results I L-1 and IL-2 of 10\+\{-3\}~10\+3 U/ml exerted no effect on basic Tg productio n . IL-1 significantly inhibited TSH-induced Tg secretion in a dose-dependent m a nner, while IL-2 increased TSH-stimulated Tg generation. Conclusion IL-1 and IL-2 act as circulating or/and local modulators of the thyroid glan ds.\;
