通腑活血法对脓毒症大鼠小肠组织的保护作用及机制研究 被引量:6

摘要 目的:探讨通腑活血法对脓毒症大鼠小肠组织的保护作用及机制。方法:雄性清洁级SD昆明大鼠100只随机分为5组,每组20只。正常对照组不作任何处理,模型组大鼠按1mL/100g/d灌胃给予生理盐水,桃核承气汤组按1mL/100g/d给予桃核承气汤药液灌胃,7d后造模。大鼠造模成功后6h、12h分别采集血清、小肠组织标本,PCR检测相关指标,电镜观察病理组织学改变。结果:①桃核承气汤组大鼠小肠组织的受损程度较模型组明显减轻(P<0.05);②与同时点模型组比较,桃核承气汤组大鼠小肠组织TLR4mRNA、12h时间点CD4mRNA表达减少(P<0.05),血清TNF-α、IL-6和LPS水平明显下降(P<0.05)。结论:桃核承气汤可减轻脓毒症大鼠小肠组织炎性损伤,其机制可能与减少CD4mRNA和TLR4mRNA表达,进而下调TNF-α、IL-6等炎症因子有关。 Objectives:To investigate the effect of Peach Kernel Purgative Decoction on TLR signal pathway and the protection of intestinal tissue of sepsis rat.Methods: Rats were randomly divided into 5 groups,including normal control group,model 6h and 12h groups,Peach Kernel Purgative Decoction 6h and 12h groups.Normal control group was not given any treatment,model group and Peach Kernel Purgative Decoction group were respectively given saline and Peach Kernel Purgative Decoction orally by 1mL/100g weight every day,Rat models were made 7 days latter.Blood and intestinal tissue were collected separately after model made 6h and 12h for normal biochemical examination and pathological dyeing.The changes of intestinal tissues were observed by electron microscopy,and the expression of TLR4mRNA and CD4mRNA were detected by PCR.Results ①the damage degree of rats'intestinal tissue in Peach Kernel Purgative Decoction group was significantly lighter than model group(P0.05);②TLR4mRNA expression in rats'intestinal tissue of Peach Kernel Purgative Decoction group was significant lower than the model group in the same time points,as well as the CD4mRNA expression in 12h group decreased significantly(P0.05);③TNF-α、IL-6 and LPS level in serum of Peach Kernel Purgative Decoction groups significant lower than the model group in the same time points(P0.05).Conclusion:Peach Kernel Purgative Decoction could reduce the inflammatory injury of sepsis rats' small intestine tissue,and the mechanism may be klated to the decrease of CD4mRNA and TLR4mRNA expression and then decrease the inflammatory factors such as TNF-α、IL-6.
