摘要
目的 研究树突状细胞 (DC)与肿瘤细胞 (SP2 /0 )融合后在体内的抗肿瘤效应。方法 体外分离诱导扩增BALB/C小鼠骨髓DC ,然后与SP2 /0细胞进行融合 ,将融合细胞接种于同基因小鼠皮下 ,观察肿瘤的生长及组织病理学特征。结果 DC与SP2 /0细胞融合后接种于小鼠能明显抑制肿瘤的生长 ,肿瘤的发生、生长速度及病理组织学改变均显示出DC SP2 /0融合细胞的抑瘤效应。结论 DC与肿瘤细胞融合后在体内可产生抗肿瘤免疫保护作用 ,有可能作为 1种新型的疫苗用于临床的肿瘤免疫治疗。
Objective To study antitumor effect in vivo induced by fusing entire tumor cells with cultured dendritic cells(DC).Methods Bone-marrow-derived DCs were isolated and induced from BALB/C mice,and expanded with GM-CSF and TNFα;then it were fused with SP2/0 cells in vitro by polyethylene glycol(PEG);DC-SP2/0 fused cells were implanted to syngeneic mice;the pathologic characterization and growth of implanted tumor were observed.Results DC-SP2/0 fused cells generated strong antitumor activity and inhibit the growth of tumor in implated mice.Conclusion Protective antitumor immunity can be induced in vivo by dendritic cells fused with tumor cells.The DC-tumor cells may become an effecfive tumor vaccine and have broad clinical application.
