摘要
目的研究白藜芦醇对戊四氮致痫大鼠的空间学习记忆能力的影响。方法采用戊四氮亚惊厥剂量腹腔注射建立慢性癫痫模型,造模成功后予以白藜芦醇15 mg/kg灌胃干预10 d,行Morris水迷宫实验测试大鼠的空间学习和记忆能力。结果经28 d连续给药,18只大鼠符合Racine点燃标准,Res干预组大鼠痫性发作潜伏期明显延长,且其发作时间明显低于癫痫模型组、二甲基亚砜组,差异有统计学意义(P<0.05)。定位航行试验:实验第1、2 d,各组大鼠逃避潜伏期差异没有统计学意义,而第3、4、5 d癫痫模型组、二甲基亚砜组大鼠逃避潜伏期比正常对照组明显延长,Res干预组大鼠逃避潜伏期明显低于癫痫模型组、二甲基亚砜组,差异有统计学意义(P<0.05)。空间探索试验:将原站台撤去后,癫痫模型组、二甲基亚砜组穿越原站台次数和象限逗留时间,比正常对照组明显减少;同Res干预组相比减少,差异有统计学意义(P<0.05)。结论白藜芦醇对戊四氮致痫大鼠具有抗癫痫和改善其空间学习记忆能力。
Objective To study the effects of resveratrol (Res) on spatial learning and memory ability of pentylenetetrazole (PTZ)-kindled model. Methods Chronic PIE-kindling model was established by injecting PTZ intraperitoneally. After kindling models were established, the Res were applied intragastrically at a dose of 15 mg/kg once a day for 10 d. The seizure latency and onset time were observed and recorded. Spatial learning and memory ability was evaluated by Morris water maze. Results After continuous dosing for 28 d, 18 rats were reaching the Racine kindling standard. Rats seizures latent period in the Res group was significantly prolonged, and its duration was significantly shorter than these of the epilepsy model group and the dimethylsulfo:dde (DMSO) group with statistically significant difference (P 〈0. 05). Place navigation test: at 1 d and 2 d, there was no significant difference in the escape latencies of four groups, while at 3 d, 4 d, and 5 d, the escape latencies of epilepsy group and DMSO group were significantly longer compared to normal control group (P 〈0.05). The escape latencies of Res group was significantly lower than those of the epilepsy model group and the DMSO group with statistically significant difference (P〈0.05). Spatial probe test: length of stay in the quadrant of the original site and the number of crossing times through the original site which removed in epilepsy model group and DMSO group were decreased obviously compared with that of normal group, while they were reduced compared with the Res intervention group with statistically significant difference (P 〈0.05). Conclusion The resveratrol plays a role in anti-epileptic, which also might improve the spatial learning and memory ability of PTZ-kindling model.
