摘要
目的:研究孕早期高蛋白膳食(HPD)对后代大鼠成年肥胖及磷酸烯醇式丙酮酸羧激酶(PEPCK)表达的影响,为肥胖的早期防治提供依据。方法:孕早期大鼠分别给予HPD和标准膳食(SD),其后代分别为高蛋白质组(HP)和对照组(CON),交叉哺乳3周后给予SD,成年后高脂膳食诱导肥胖。分阶段处死大鼠取白色脂肪,荧光定量PCR法检测PEPCKmRNA的水平。结果:与SD大鼠后代相比,HPD大鼠后代的体重降低(P<0.05),PEPCK mRNA水平下降。结论:孕早期HPD可程序性降低后代PEPCK mRNA的水平,减少了成年肥胖的发生。
Objective: To explore the effects of high protein diet during early pregnancy on offspring phosphoenolpyruvate car- boxykinase (PEPCK) gene, and find out the method of preventing obesity. Methods: Rats were received a high protein diet (HPD) and standard diet (SD) during early pregnancy, respectively. Their offspring were divided into high protein group (HP) and control group ( CON), respectively. Offspring were fed with SD after weaning. At adult, rats were fed with high fat diet (HFD) to induce obesity. At dif- ferent stages, offspring were sacrificed for analyzing PEPCK gene level. Results: Body weight and PEPCK gene level of offspring of HPD during pregnancy were decreased vs offspring of SD during pregnancy ( P 〈 0.05 ) . Conclusion: HPD during early pregnancy can prevent offspring from obesity and decrease PEPCK gene level.
