肝细胞肝癌中RUNX3和Galectin-3的表达及意义

Expression and clinical significance of RUNX3 and Galectin-3 in hepatocellular carcinoma tissues

摘要 目的探讨肝细胞肝癌(HCC)中RUNX3和Galectin-3的表达及其临床意义。方法采用逆转录聚合酶链反应(RT—PCR)和蛋白质印迹(Westernblot)的方法检测RUNX3和Galectin-3在34例HCC组织、28例癌旁组织和18例正常肝组织组织中的表达,并结合临床病理指标进行分析。结果RUNX3的mRNA及蛋白在HCC组织中的表达显著低于癌旁及正常对照组织(P〈0.05);Galectin-3的mRNA及蛋白在正常对照组织中失表达,在HCC组织中的表达高于癌旁组织,差异有统计学意义(P〈0.05)。两者在HCC组织中的表达呈负相关(r=-0.247,P=0.019)。结论RUNX3和Galectin-3基因在HCC的发生中起重要作用,在HCC组织中RUNX3蛋白的低表达和Galectin-3蛋白的高表达可能共同促进了HCC的发生和发展。 Objective To investigate the expression and clinical significance of RUNX3 and Galactin-3 in henatocellular carcinoma(HCC) tissues. Methods The expression of RUNX3 and Galectin-3 in 34 liver carcinoma, 28 paracarcinoma tissues and 18 normal liver tissues were detected by Western blot, RT-PCR and the result was ana lyzed combined with the clincopathlogical indexes. Results The expression of RUNX3 mRNA and protein in hepatocellular carcinoma were lower than those in paracarcinoma tissues and normal comparison (P 〈 0. 05 ) ; The expression of Galectin-3 mRNA and protein in nomal comparison was absence. The expression of Galectin-3 in HCC was higher than that in paracarcinoma, there were significant drfferences ( P 〈 0. 05 ) ; Negative correlation was found between the positive expression rate of RUNX3 protein and Galectin-3 protein in HCC( r = -0.247, P = 0.019). Conclusions RUNX3 and Galectin-3 play an important role in the development of HCC. The low expression of RUNX3 protein and high expression of Galectin-3 protein may promote the malignancy change of HCC.
