商品化口蹄疫疫苗与猪瘟疫苗对小鼠免疫效力的研究 被引量:1

Study of immune efficacy on co-immunization of two commercial vaccines in BALB/c mice

摘要 通过研究口蹄疫疫苗和猪瘟疫苗在小鼠模型上免疫后的免疫效力变化,以便探讨共免疫两种疫苗与单独免疫其中一种疫苗对小鼠免疫应答作用的差异。将小鼠分成以下几个免疫组进行接种或注射:①口蹄疫疫苗单免组;②猪瘟疫苗单免组;③口蹄疫疫苗与猪瘟疫苗共免组;④PBS注射组。二免后第7天采集血液,分离血清,检测抗体效价及血清中IFN-γ、IL-4的质量浓度;二免后第7天分离小鼠脾淋巴细胞,分别进行淋巴细胞增殖试验及对脾淋巴细胞CD3+CD4+和CD3+CD8+T淋巴细胞亚群分化进行检测。结果,共免疫两种疫苗的小鼠产生的口蹄疫抗体效价比单独免疫口蹄疫疫苗的要低,但是产生的猪瘟抗体效价比单独免疫猪瘟疫苗的要高很多;共免组产生的INF-γ比单免口蹄疫疫苗组的要低,但是比单免猪瘟疫苗组的要高。淋巴细胞增殖试验及流式细胞仪检测的结果表明,共免疫口蹄疫疫苗和猪瘟疫苗对小鼠脾淋巴细胞再次识别特定抗原的应答能力及T细胞亚群分化均具有重要影响。结果表明,共免疫这两种疫苗对小鼠体液免疫反应影响较小,甚至有促进作用,但是细胞免疫应答受到了抑制。 In order to detect the immune efficacy of simultaneous vaccination using two commercial vaccines against foot-and-mouth disease(FMD) and classical swine fever(CSF) in a mouse model, the immune responses after immunity was studied. The BALB/c mice were divided into four groups: (1)FMDV vaccine alone; (2)CSFV vaccine alone; (3)FMDV vaccine+CSFV vaccine; (4) PBS. The antibody titers,IFN-γ, and IL-4 in the serum were detected on day 7 after the second vaccination. The lymphocyte prolifera- tion test and CD3+ CD4+/ CD3+ CDS+T cells detection was performed using the splenic lymphocytes isolated on day 7 after the second vaccination. The antibody titer against FMD in the co-immune group was lower than that of the FMD vaccination group,but the antibody titer against CSF in the co-immune group was higher than that of the CSF vaccination group. The level of INF-γ of the co-immune group was higher than that of the FMD vaccination group, but lower than that of the CSF vaccination group. Through the detection of the proliferation of splenic lymphocytes of mice and the flow cytometry analysis, we found that,co-immunization had an important influence on the immune response. In conclusion, this study suggests that the effective humoral immunity responses were induced by co-immunization of FMD vaccine and CSF vaccine. However,the cellular immune responses to FMDV were inhibited by co-immunization.
